A STUDY OF CLINICAL APPLICATION OF CULTURED EPITHELIAL AUTOGRAFT



Abstract
With recent development of cell culture, a small piece of epithelium can generate large amounts of cultured epithelium, A 2-3cm2 sized full thickness skin sample was excised from the inguinal region. and was cultured with NIH 3T3 cells treated with mitomycin C. The cultured epithelia were grafted to the donor site of split thickness skin graft and abraded wound. And the biopsy specimens were taken from the grafted area of cultured epithelia at various stages.
This study was undertaken to investigate the human epithelial cultivation and its clinical application.
The results were as follows ;
1) A 176cm2 sized cultured epidermal sheet could be obtained from 2-3cm2 of healthy skin.
2) The grafted sites healed completely within 1 week and the epithelialized area with cultured graft could be observed.
3) The cultured epithelia showed normal differentiation and developed a horny layer after 1 week. The collagen fiber was observed in basement membrane. Each layer of epithelium was thickened and the collagen fiber on loose dermoepidermal junction were noted. Clear cells, melanotic granule and melanocyte were noted after 2 weeks. Rete ridge became prominent and dendritic process of melanocyte was observed after 3 reeks. After 4 weeks, nearly normal epidermis was noted.
This report presents our experience with cultured epithelial autografts. It can be concluded that cultured human epithelium was taken well and normal differentiation was noted at 4 weeks after grafting.
